Abstract: OBJECTIVE: To characterize associated receptors of functionality in melanocytes expanded ex vivo in selective media in two times of the culture.METHODOLOGY: We conducted a quasi-experimental study with basic tissue obtained by biopsy of skin of healthy individuals over the age of 18 who participated voluntarily. The sample size was of convenience with a total of 15 participants. The tissue obtained from the first 10 was used for the standardization of the culture. With the tissue obtained from the last 5 the survey was conducted taking the culture to confluence of 80% - 90% in the second pass. The major variables measured were: 1. The number of melanocytes present in each pass, and the expression of receptors of functionality (c-kit, Melan A and HMB45) in these cells. RESULTS: The tissue was obtained from 5 individuals in the masculine gender with an average age of 26.8 years and phototype II, III and IV. After the process of obtaining and cultivation in selective media are achieving the successful growth of melanocytes ex vivo with a mean time to achieve confluence of 80% to 90% on the first pass of 35 days and for the second pass of 51.2 days. The average number of melanocytes obtained by culture in the first pass was 233,000 cells and for the second pass was 553,000 cells. The average number of melanocytes observed by mm2 with hematoxylin and eosin in tissue was 6.4 .The average expression of the HMB45 receiver on the first pass was 9 %, for Melan - 30% and for C- Kit 9 %. In the second pass the expression of HM45 was 62 %, for Melan TO 81% and for C- Kit of 37 % There was an increase of the percentage of expression of the receptors of functionality between the first and second pass with an average of 53% increase in HMB45, an average of 51% for Melan A and an average of 28% increase for c- kit. CONCLUSION: The advanced cell therapy with cultivation of melanocytes opens the door to a therapeutic alternative with optimal conditions to obtain viable and functional melanocytes to ensure their best implantation in patients with vitiligo. This implantation given the results obtained would be ideal in the second pass of the culture where melanocytes increase their expression of receptors of functionality which would herald the introduction and successful repigmentation.
